Moderna Cancer Vaccine Stops Melanoma Returning: What’s Next for Personalized Treatments?

The companies said the vaccine cut the risk of recurrence by 50% and spread by 59% in a prior mid-stage trial.

  • On Wednesday, Moderna and Merck announced their personalized mRNA vaccine helped prevent melanoma recurrence in a large trial involving more than 1,000 patients who had localized tumors surgically removed.
  • Their treatment, intismeran autogene, combines a customized mRNA vaccine with Merck's immunotherapy drug Keytruda to train the immune system to specifically target mutations found only on a patient's tumor cells.
  • The study enrolled around 1,100 people with advanced melanoma; previous mid-stage trial data showed the vaccine cut the risk of cancer returning by half and reduced spread risk by 59 per cent compared with Keytruda alone.
  • Dr. Julie Gralow, chief medical officer of the American Society of Clinical Oncology, called the results "a monumental leap forward," while Dr. Elizabeth Buchbinder, a melanoma specialist at the Mass General Brigham Cancer Institute, said it "may change that math" for treatment.
  • Researchers are investigating potential applications for other cancers, with colon cancer trial results expected in 2027 and pancreatic cancer trials following in 2031.
